Prostate Cancer 2: A Comparative Study of Histochemical Stains in Prostate Cancer

In a retrospective study, the effectiveness of Masson trichrome and VanGieson stains in highlighting collagen fibers in prostate cancer tissues was examined. A total of thirty specimens from 2015-2018 were randomly selected, and stained with both stains. Patient ages were categorized, with the majority falling within the 50-90 years group. VanGieson demonstrated higher intensity for muscle and RBC staining, with 66.7% of cases showing the highest intensity. Meanwhile, Masson trichrome showed a range of intensities, with 43.3% of cases exhibiting the second-highest intensity.

The study found that VanGieson stain outperformed Masson trichrome in highlighting muscles and collagen, a result consistent with existing literature. VanGieson's ability to vividly and distinctly stain muscle, collagen, and RBCs underscored its superiority. In contrast, Masson trichrome, while sensitive, required time-consuming preparation of staining solution and individual differentiation of slides, impacting the final outcome. Notably, the chance of over-differentiation with acetic acid was higher, potentially leading to staining disappearance.

Based on the findings, the study recommends using larger sample sizes for more robust results and employing advanced techniques like immunohistochemistry and tumour markers for enhanced reliability. Additionally, emphasizing the importance of differentiation and vigilance against stain fading or disappearance is advised for staff handling such specimens.
